<P>PROBLEM TO BE SOLVED: To provide modified vitamin K-dependent polypeptides and methods of modulating clot formation in mammals. <P>SOLUTION: The invention provides the vitamin K-dependent polypeptides with enhanced membrane binding affinity. The modification in vitamin K-dependent γ-carboxyglutamic acid (GLA) domain enhances membrane binding affinity of the polypeptide. The modified vitamin K-dependent polypeptide has enhanced membrane activity, and can be used as anti-coagulants, pro-coagulants, or for other functions that utilize vitamin K-dependent proteins. For example, an improved factor VII molecule may provide several benefits by lowering the dosage of VIIa needed, reducing the relative frequency of administration, and/or by providing qualitative changes that allow more effective treatment of deficiency states.
