PROBLEM TO BE SOLVED: To provide an agent for promoting formation of root nodules that can be easily applied to soil or plants and can efficiently form root nodules, and to provide a method for promoting formation of root nodules using the agent. SOLUTION: The agent for promoting formation of root nodules containing as the active ingredient a compound selected from the group consisting of a nucleoside, a nucleotide and a nucleic-acid base, for example, inosine, is applied to leguminous plants to promote formation of the root nodules.
