<P>PROBLEM TO BE SOLVED: To provide a method for amplifying a nucleic acid sequence enabling the amplification of a specific nucleic acid sequence by a simple procedure compared with conventional method and a method for detecting the amplification product. <P>SOLUTION: The method for the amplification of a nucleic acid sequence contains (1) a step to bring (a) a specimen presumed to contain a target nucleic acid sequence, (b) a target nucleic acid sequence capturing probe containing a complementary sequence annealing to the 3'-terminal region of the target nucleic acid sequence, a promoter sequence recognizing an RNA polymerase and one of an affinity molecule pair and (c) a carrier produced by fixing the other member of the affinity molecule pair on a fine particle to mutual contact and form a complex composed of the target nucleic acid sequence, the target nucleic acid sequence capturing probe and the carrier when the target nucleic acid sequence is present in the specimen and (2) a step to perform a nucleic acid sequence amplification reaction by using the target nucleic acid sequence as a template in the presence of a reverse transcriptase having RN-ase H activity and an RNA polymerase or in the presence of a reverse transcriptase, an RN-ase H and an RNA polymerase when the target nucleic acid sequence is present in the specimen. <P>COPYRIGHT: (C)2006,JPO&NCIPI |
