PURPOSE: A method for detecting metagenome library-derived enzyme activation is provided to be applied to protein engineering. CONSTITUTION: A method for detecting metagenome library-derived enzyme activation comprises: a step of preparing environment-derived metagenome library; a step of preparing a re-constructed gene circuit for sensing phenol chemicals; a step of introducing the metagenome library and re-constructed gene circuit to a host microbe to obtain transformed microorganism library; a step of treating the microbe with phenol release compound; a step of measuring reporter protein activity; and a step of collecting enzyme gene and identifying by sequencing.
