<p>The method for sequencing C-terminus of proteins, comprises transforming free COOH-group in protein with marker A, removing free marker A, splitting the marked protein in peptide, transferring newly available COOH-group if necessary with marker B that is different from marker A, identifying C-terminus of peptide piece marking with the marker A, and sequencing the C-terminus of peptide piece in usual manner marked with the marker A. The coupling of free COOH-groups is carried out with the marker A and B after activation of the COOH-groups. Carbodiimide if necessary in combination with N-hydroxysuccinimide is used for the activation. A marker-coupled activation reagent is used for the activation of the free COOH-groups. Fluorescent dyes are used as marker. Enzyme is used for splitting of protein. The enzyme is trypsin or V8 and is immobilized on a stationary phase.</p> |
