PURPOSE: A method for detecting methylation of a breast cancer-specific methylated marker is provided to quickly and accurately detect methylation. CONSTITUTION: A method for methylation of a breast cancer-specific methylated marker comprises: a step of preparing a clinical sample containing DNA; a step of detecting CpG island methylation of OTP(NM_032109, Orthopedia Homeobox) from the DNA. The CpG island methylation is detected at an intron site of OTP gene. The intron of OTP gene has a base sequence of sequence number 1. A composition for diagnosing breast cancer contains GpG island of OTP gene.
