<p>The invention relates to a process for protectingα,ωdiamino acids as urethanes to be employed in the synthesis of certain polypeptides. According to the invention, the process consists in selectively introducing the protective groups in the two amino functional groups by simultaneously blocking the carboxyl and theα-amino group by using copper sulphate and sodium carbonate for the control of the pH value in the range of 8.5...10.5, after which the resulting copper complex is treated with tert-butyl bicarbonate with a view to protecting theω-amino group, the copper complex is split with citric acid by salification with NaCl and the precipitate is filtered and washed, then is dissolved into a 10% potassium carbonate solution and is treated with a solution of fluorenyl-methyloxycarbonyl-O-succinimide at a pH value of 8.5...10.5, after the reaction end and the adequate processing, there resulting the selectively protected product.</p> |
