Heading C2C. 5<SP>1</SP>-Inosinic acid, inosine or a mixture thereof is obtained by inoculating a mutant induced from a micro-organism of the genus Bacillus which requires both (1) adenine and (2) guanine or xanthine for its growth, on to a culture medium containing both an adenine source and either a guanine source or a xanthine source, incubating the culture medium to accumulate therein the desired substance, and recovering the desired substance from the culture medium. The adenine source may be adenine itself, a compound which contains an adenine component in its molecule and is easily convertible into adenine or a natural substance containing adenine; the guanine source may be guanine itself, a compound readily converted into guanine or a natural substance containing guanine and the xanthine source may be xanthine itself or a derivative of it. Natural substances containing both adenine and guanine may also be used. The culture medium may also contain an assimilatable carbon source, a digestible nitrogen source, a small quantity of inorganic salts and other conventional nutrient factors, e.g. vitamins, and the initial pH of the culture medium and the incubation temperature may be 6À0-8À0 and 20- 40‹ C. respectively. Specified mutants are those induced from Bacillus subtilis Cohn emend Prazmowski, Bacillus megaterium de Bary and Bacillus frumilus Gottheil by treating them with ultra-violet radiation, X-rays, nitrogen mustard or nitrous acid. Specified adenine sources are adenine, adenosine, 3<SP>1</SP>- adenylic acid, succinoadenylic acid, meat extract, cornsteep liquor and yeast extract, specified guanine sources are guanine, guanosine, 3<SP>1</SP>(5<SP>1</SP>)-guanylic acid, meat extract, cornsteep liquor and yeast extract, specified xanthine sources are xanthine, xanthosine and 3<SP>1</SP>(5<SP>1</SP>)-xanthylic acid and specified sources of both adenine and guanine are meat extract and yeast extract.
