PURPOSE: A visible SALDI mass spectrometry of biomolecules using protonation agent is provided to enable a sample to be effectively analyzed by selecting a substrate efficient to light absorbing and energy transfer and enhancing the efficiency of the ionization of the sample. CONSTITUTION: A visible SALDI mass spectrometry of biomolecules using protonation agent is as follows. A sample and protonation agent are adsorbed on a substrate. Laser is irradiated to the adsorbed sample. The substrate is a carbon substrate. In the adsorbing step, the protonation agent is adsorbed after the sample is adsorbed. The protonation agent includes glycerol. The used laser is visible laser. The sample is biomolecule.
