SIRNA SEQUENCES TARGETING ENDOGENOUS PRION PROTEIN OF MOUSE NEUROBLASTOMA AND BOVINE SPONGIFORM ENCEPHALOPATHY SPECIFIC CELL CULTURE MODEL

PURPOSE: A method or manufacturing a cell model which is specific to BSE(bovine spongiform encephalopathy) is provided to understand the disease and to develop a biomarker for preventing and treating BSE. CONSTITUTION: An siRNA is denoted by sequence number 2. The siRNA suppresses mouse prion gene(sequence number 3, Genebank Accession No.NM_011170). A method for constructing a cell model which is specific to BSE(bovine spongiform encephalopathy) comprises: a step of cloning a bovine prion gene(boPrnp ORF, sequence number 4) into an eukaryotic cell expression vector; a step of transplanting the vector to mouse nervous cell line; and a step of transducing siRNA of sequence number 2 to the mouse nervous cell line to suppress expression of mouse prion protein.
