PURPOSE: A composition for preventing and treating TNF-alpha-mediated diseases is provided to suppress ARE(AU-rich element) binding with TNF-alpha mRNA. CONSTITUTION: A composition for preventing and treating TNF-alpha-mediated diseases contains a compound of chemical formula I or pharmaceutically acceptable salt threof as an active ingredient. The compound of chemical formula I is 2-amino-4-(4-(benzyloxy)phenyl)-6-(2-(3,4-dehydroxyphenyl)-2-oxoemthylthio)pyridine-3,5-dicarbon nitrile or 2-(6-amino-3,5-dicyano-4-(4-methoxyphenyl)pyridine-2-ylthio)-N-(4-chlorophenyl)aceteamide. The TNF-alpha-mediated diseases are sepsis, bone diseases, hepatitis, alcoholic hepatitis, viral hepatitis, coagulation, burn, reperfusion injury, or fever.
