The present case relates to ClfA polypeptides wherein amino acid Y474 or an amino acid adjacent to Y474 is mutated such that fibrinogen binding activity is decreased compared to an equivalent ClfA polypeptide without mutation of Y474 or an amino acid adjacent to Y474. Immunogenic compositions, vaccines, processes and uses of such mutated ClfA polypeptides in the treatment of staphylococcal disease are also claimed.
