PURPOSE: A medium composition which is effective in proliferating and maintaining pluripotent stem cells is provided to effectively culture the stem cells. CONSTITUTION: A medium composition for maintaining and culturing undifferentiated pluripotent stem cells contains 0.01-100 uM of neuropepetide Y. The pluripotent stem cells are embryonic stem cells or induced pluripotent stem cells. The pluripotent stem cells are derived from human. A method for preparing pluripotent stem cell line comprises: a step of collecting pluripotent stem cells; and a step of culturing the stem cells under the presence of the medium composition.
