A radio frequency identification system and an operation method thereof. The tag states of the radio frequency identification system comprises a power-off state, an initialization state, an inventory state, an acknowledged state, an access state and a killed state, and the operation method of the radio frequency identification system includes the following steps: a reader inventories the tag to obtain a random number generated by the tag and the tag ID of the tag; the reader accesses the tag according to the random number or the tag ID, which includes: the reader accesses the tag through a direct access command with the tag ID when the tag is in the initialization state, the inventory state or the acknowledged state; the reader accesses the tag through the direct access command with the tag ID or through a common access command with the random number when the tag is in the access state. The system and method can ensure the tag states transfer normally and the situation of a state machine endless loop does not occur, and a more flexible access interface can also be provide , thus the access efficiency is improved.
