PROBLEM TO BE SOLVED: To provide a reagent, method and kit for purification of RNA from a biological sample. SOLUTION: There are provided a formulation for isolating and purifying nucleic acids comprising a lithium salt at concentration of at least about 1 M, a surfactant, and a buffer. The invention provides a formulation for isolating and purifying nucleic acids, in one embodiment, comprising at least a lithium salt at concentration of at least about 1 M, a surfactant, a buffer and if necessary a chelating agent, if necessary a reducing agent, and if necessary a tungstate, wherein the solution has a pH of above about 7.
