<P>PROBLEM TO BE SOLVED: To provide a useful and safe viral vector. <P>SOLUTION: There is provided a multicistronic retroviral vector genome comprising a first nucleotide sequence upstream of an internal regulatory element, such that the level of genomic RNA available for packaging in the absence of rev, or a functional equivalent thereof, is increased. There is also provided a vector genome comprising a nucleotide sequence which is operably linked to a viral LTR and is upstream of an internal regulatory element.
