Disclosed is the use of an imine compound of formula (I) or (VII) or a pharmaceutically acceptable derivative thereof as hereinbefore defined, wherein X is O, S, or NR, where R and the further substituents are defined in the specification in the manufacture of a medicament for inhibiting or preventing alpha-synuclein toxicity and/or fibril formation, inhibiting or preventing a-synuclein fibril growth, and/or causing disassembly, disruption, and/or disaggregation of a-synuclein fibrils and alpha-synuclein-associated protein deposits. Also disclosed is the use of said compound of in the manufacture of a medicament for treating or ameliorating one or more symptoms of a synuclein disease, synucleinopathy or synuclein toxicity e.g. Parkinson's disease, a composition comprising said compound, a method of inhibiting or preventing alpha-synuclein toxicity and/or fibril formation, inhibiting or preventing a-synuclein fibril growth, and/or causing disassembly, disruption, and/or disaggregation of a-synuclein fibrils and alpha-synuclein-associated protein deposits in an isolated cell comprising contacting the cell with the said compound and said compound.
