Disclosed is a method for preparing a purine derivative of formula (I), where the substituents are as defined in the specification, which is: reacting a compound of formula (VI) with a compound X-R3-X, where X is a leaving group, to obtain a compound of formula (V), reacting compound (V) with CH2(COOR4)2 to obtain a compound of formula (IV), reducing compound (IV) to obtain a compound of formula (III), hydrogenating compound (III) to obtain a compound of formula (II), and reacting compound II with (R5)2O to obtain compound (I). Specific solvents and other reaction conditions are also described, as are suitable substituents for making famiciclovir.
