PROBLEM TO BE SOLVED: To provide a method for determining a brightness value of a target region on an optical image. SOLUTION: The method for determining the target brightness value of a target region for an immunological analyzer, includes the steps of photographing an object to be photographed containing a background region and a target region by a camera, providing the image of the object to the camera, converting the image by an image sensor to an electrical signal, determining a quadratic approximation equation y=ax<SP>2</SP>with x indicating a position value in the object, y indicating a brightness value with x, and an indicating a coefficient, determining the difference between y and the brightness value at the zero point (the origin), correcting y caused by uneven illumination and/or lens bending using the difference, and determining the target brightness value of the target region with a brightness value y' that is corrected by the x and the y corrected.
