<P>PROBLEM TO BE SOLVED: To provide determining materials for variety identification in konjac (devil's tongue jelly) industry by constructing a DNA marker detecting polymorphisms among the varieties of konjac. <P>SOLUTION: By subjecting 4 main cultivated varieties (Akagioodama, Harunakuro, Myougiyutaka and Miyamamasari) and other one lineage (a Chinese variety) under a restricting enzyme landmark genome scanning (RLGS), DNA fragments specific to these varieties and lineage are detected among the genome DNAs of them. From the base sequence information of the detected DNA fragments, a primer specifically amplifying the DNA fragments showing the polymorphism among these varieties and lineage is constructed. As a result of trying PCR reactions by using the primer, it is elucidated that the identification of the main 4 cultivated varieties of the konjac is possible.
