A method for culturing a human embryonic stem cell to form undifferentiated colony from an embryonic stem cell is provided to increase the rated of undifferentiated colony formation and massively culture human embryonic stem cell. A method for culturing a human embryonic stem cell comprises: a step of culturing the human embryonic stem cell group in a medium containing an inhibitor of Rho GTPase regulator; a step of isolating the human embryonic stem cell group into monocell using cell connection weakening enzyme; and a step of further culturing the human embryonic stem cell. Rho GTPase is Rho, Rac or Cdc42. Rho GTPase regulator is ROCK, PKN/PRK1, PRK2, mDia, PI5-kinase, IQGAP1, WAVE complex, PI3-kinase, PAK, Por1, p140Sra-1, POSH, MLK, NADPH oxidase, WASP/N-WASP, Ack, PAR-6, or Cotamer. |
