Disclosed is a novel means for amplifying a target gene outside the chromosome of a mammalian cell at a higher ratio in the amplification of the target gene using an IR/MAR plasmid, for the purpose of further improving a high-level gene amplification system. Specifically disclosed is a method for amplifying a target gene outside the chromosome of a mammalian cell, which is characterized by involving a gene introduction step of introducing a vector, the target gene and a polynucleotide comprising a telomere repeat sequence simultaneously into the mammalian cell, wherein the vector comprises a mammalian replication initiation region that can act in a mammalian cell and a nuclear matrix binding region that can act in a mammalian cell.
