PROBLEM TO BE SOLVED: To provide a method for easily and inexpensively assaying helicase enzyme activity, and to provide a reagent therefor. SOLUTION: The method for assaying the helicase activity assays the followings: a partially double-stranded nucleic acid (A) comprising a single-stranded nucleic acid (A-1) comprising a sequence including, on the 5'- and/or 3'-end, a sequence (helicase recognition sequence) necessary for the enzymatic reaction by helicase and a single-stranded nucleic acid (A-2) comprising a sequence complementary to the moiety other than the helicase recognition sequence among the (A-1) nucleic acid; a single-stranded nucleic acid (B) comprising a sequence partially or wholly complementary to the nucleic acid (A-2) and comprising a sequence including no helicase recognition sequence when formed a base pair with the nucleic acid (A-2); and a thus formed double-stranded nucleic acid or partially double-stranded nucleic acid (C) obtained by reaction in coexistence of helicase, and comprising the nucleic acid (A-2) and the single-stranded nucleic acid (B).
