The present invention relates to a buffer composition for catalyzing the preparation of calcitriol or calcifediol and a method for preparing calcitriol or calcifediol using the same and more specifically to a buffer composition for catalyzing the production of calcitriol or calcifediol comprising a metal compound, an organic solvent, cyclodextrin, tris(hydroxymethyl)aminomethane, sodium succinate, sodium chloride, magnesium chloride and water, and to a method for preparing calcitriol or calcifediol using the same. The production method of the present invention affords high preparation yields of calcitriol or calcifediol and carries out bioconversion not in a microbiological culture system but in an enzymatic reaction system, and thus does not require aseptic conditions. Because isolation and purification is initiated under cleaner conditions than those used for the microbiological culture method upon completion of a biocatalytic reaction, this method is advantageous in that the cost of separation is low and the quality is superior. In addition, the buffer composition of the present invention for catalyzing the preparation of calcitriol or calcifediol effectively provides superior productivity of calcitriol or calcifediol.
