The present invention relates to: a p53 recombinant protein having cell permeability by fusion of a macromolecule transduction domain (MTD) with a tumor suppressor p53, a polynucleotide encoding the cell permeable p53 recombinant protein, an expression vector of the cell permeable p53 recombinant protein, and an anti-cancer pharmaceutical composition containing the cell permeable p53 recombinant protein as an active ingredient. According to the present invention, the cell permeable p53 recombinant protein effectively reactivates the mechanism of signal transduction relating to cell cycle suppression and apoptosis induction by transferring tumor suppressor p53 into the nucleus of a cell and suppressing formation of cyclin-dependent kinase and cyclin complex. Therefore, a composition containing the cell permeable p53 recombinant protein as an active ingredient can be used as an anti-cancer drug for preventing and/or treating various cancers.
