<p>PURPOSE: A microbiological method for monitoring environmental effect of acid mine drainage is provided to monitor mine drainage by detecting microorganisms. CONSTITUTION: A method for monitoring environmental effect of acid mine drainage by real time PCR comprises: a step of collecting river or mine drainage(sample) and storing at 4°C; a step of centrifuging the sample and concentrating to separate cells(S120); a step of extracting genomic DNA from precipitate(S130); a step of amplifying DNA by PCR to produce standard DNA from the genomic DNA(S140); and a step of purifying PCR product and quanlifying(S150). The precipitate contains Ferribacterium limineticum, Desulfosporosinus orientus, Acidithiobacillus ferrooxidans, Leptosprillum ferrooxidans, or Acidiphilium cryptum JF-5.</p> |
