PURPOSE: A method for manufacturing glycerol mono-t-butyl ether is provided to maximize the conversion of glycerol and the selectivity of the glycerol mono-t-butyl ether by controlling reaction terminal time and to optimize the yield of the glycerol mono-t-butyl ether. CONSTITUTION: A method for manufacturing glycerol mono-t-butyl ether includes a step of manufacturing the glycerol mono-t-butyl ether by reacting glycerol or a composition containing the glycerol with liquid isobutene in a hydrophobic solvent using a catalyst for etherification. The hydrophobic solvent is an ether-based solvent. The glycerol or the composition containing the glycerol is obtained from by-products of a biodiesel production reaction.
