PURPOSE: A biosensor for detecting histidine and protein containing a large amount of histidine is provided to simply and conveniently detect histidine. CONSTITUTION: A histidine-specific biosensor comprises: a metal particle and organic ligand which is conjugated on the surface of metal particle. The organic ligand has a carboxylic group. The metal particle is silver, gold, or platinum. The diameter of the metal particle is 1nm-30nm. The organic ligand is a lysine with carboxylic group. A method for detecting histidine using the biosensor comprises: a step of dispersing the biosensor in a detection buffer; a step of mixing histidine-containing solution with the buffer; and a step of observing color change of the detection buffer. The biosensor is used for diagnosing liver cirrhosis, AIDS, asthma, lung disease, thrombotic disorder or malaria.
