METHOD FOR DETERMINATION OF ACTIVITY OF BIOLOGICALLY ACTIVE SUBSTANCE

Method for determination of activity of biologically active substance (BAS) prescribes investigation of effect of BAS on growth of somatic cells of Drosophila melanogaster Meig. First one prepares nutrient medium with addition of BAS with given (experiment) and zero (control) concentrations. After that on nutrient medium prepared one grows larvae of drosophila. At the end of the 3-d stage of development in larvae salivary glands are separated, from those one prepares pressed acetoorceinic preparations of polytenic chromosomes. After that on those preparations one determines percent distribution of niclei with different stage of polyteny of chromosomes (SPC). After that one calculates average values of SPC. By difference between average values of SPC in experiment and control one makes conclusion on activity of BAS.
