A method for cryopreservation of testicular cell suspension relates to cryobiology and cryomedicine and can be used in practical medicine, in particular in transplantology. According to the method the cell suspension is combined with a solution of cryoprotector DMSO in the medium of DMEM/F 12 with addition of 20 mM Hepes, taken in concentration of 10 %, and programmed freezing is conducted from temperature of 20...22 °C to the crystallization temperature at the rate of 1...2 °C/min., than programmed release from supercooling is carried out, and than the cooling rate is increased to 15...20 °C/min., cooling is performed to the temperature of -40 °C, and in the range from -40 °C to -70 °C cooling is made at the rate of 20...25 °C/min., than samples are submerged in liquid nitrogen (-196 °C). |
