<p>A circular interfering RNA (ciRNA) molecule comprising the sense and the antisense strands of an interfering RNA molecule targeting a gene of interest, wherein said sense and antisense strands are closed at one end by a loop structure and at the other end by a splice junction sequence generated by splicing activity of permuted intron-exon sequences, and wherein the circular RNA molecule inhibits the expression of the gene of interest in cells expressing said gene of interest. Genetic constructs and transcription vectors for the production of said ciRNA. Method for producing large amounts of said ciRNA in yeast mitochondria in a form which is stable and easy to purify.</p> |
