METHOD FOR PREPARING ANTI-IDIOTYPE ANTIBODY

PROBLEM TO BE SOLVED: To provide a method for producing and identifying a molecule mimicking the one idiotype of autoimmune disease-associated autoantibody.

SOLUTION: This method comprises (a) to purify an autoantibody from serum of a patient of an autoimmune disease; (b) to form an affinity matrix by bonding the autoantibody to a solid phase; (c) to make a pooled plasma or B cells comprising immunoglobulins in contact with the affinity matrix and then remove un-bound plasma components; (d) to elute bound immunoglobulin which is the anti-idiotype antibody (anti-Id) against the autoantibody from the matrix; (e) to provide a molecular library containing a plurality of molecular members; and (f) to make the anti-Id in contact with the molecular library, and isolate the bound molecules bound with the anti-Id (the bound molecules are mimicking molecules of the one idiotype of the autoantibody).
