A separation device such as a chromatography column (30) comprises a separation chamber (203) having a housing (202) for a stationary phase (204). The separation device further comprises a filling port (315) for filling the stationary phase (204) through a filling channel (310) into the separation chamber (203), and a locking piece (300; 400; 500; 600; 700) comprising the filling channel (310). The locking piece is configured to move the filling channel (310) from a first position (Figs. 3A, 3B; 4A, 4B, 4D; 5B, 5C, 5D; 6A; 7A, 7B), wherein an opening of the filling channel (310) opens into the separation chamber (203), to a second position (Figs. 3C, 3F; 4E; 5E; 6B), wherein the separation chamber (203) is locked against the filling channel (310). The locking piece (300; 400; 500; 600; 700) is configured to move the opening of the filling channel (310) substantially laterally with respect to the housing (202).
