<P>PROBLEM TO BE SOLVED: To provide a DNA amplification method that does not require a plurality of temperature settings in a series of processes from a pretreatment process of blood sample not requiring a DNA purification to a DNA amplification reaction process. <P>SOLUTION: The method for amplification of DNA from a blood sample comprises: a pretreatment process for treating a blood sample having a DNA to be amplified with an alkali aqueous solution at a normal temperature so as to extract a double-stranded DNA from the blood specimen and dissociate the double-stranded DNA into a single-stranded DNA to obtain a blood-derived sample including the single-stranded DNA; a process for mixing the blood-derived sample with at least a primer, dNTP, a strand-displacing DNA polymerase, a magnesium salt and a buffer to prepare an isothermal amplification reaction solution and constructing the isothermal amplification reaction system meeting optimum conditions for the strand-displacing DNA polymerase; and an isothermal amplification process for amplifying DNA in the isothermal amplification reaction system using the single-stranded DNA as a template. <P>COPYRIGHT: (C)2010,JPO&INPIT |
