Provided herein are methods of assessing responsiveness to an agent, methods of customizing treatments, methods of screening agents, and methods of treatment. Also provided are systems and kits for carrying out the methods contemplated herein. An exemplary aspect includes a method of assessing whether a subject having a condition, disorder, dysfunction, or disease would respond to treatment with an agent. The method comprises obtaining a stem cell sample from the subject; culturing the sample; exposing the sample to an agent; evaluating the sample in the presence of and in the absence of the agent to create a sample profile; and comparing the sample profile to a reference profile indicative of responsiveness to the agent. The similarity between the sample profile and the reference profile indicates the subject's responsiveness to the agent.
