POLYMERASE DERIVED FROM THERMOCOCCUS MARINUS AND ITS USE

PURPOSE: A DNA polymerase derived from Thermococcus marinus is provided to use in nucleic acid polymerization such as PCR. CONSTITUTION: A heat resistant Tma DNA polymerase isolated from Thermococcus marinus has an amion acid sequence of sequence number 12. The Tma DNA polymerase is optimally active at pH 7.0, 75°C, 0-20 mM of KCl, 0-15 mM of (NH_4)_2SO_4, and 12-16 mM of Mg^2+. A nucleic acid encoding the Tma DNA polymerase has nucleic acid sequence of sequence number 11. A method for manufacturing Tma DNA polymerase comprises: a step of preparing a vector expressing Tma DNA polymerase; a step of transforming the vector to microorganism; a step of culturing the transformant; and a step of collecting the proteins from the transformant.
