In the present invention, there is disclosed a human monoclonal antibody or antigen-binding portion thereof that specifically binds to insulin-like growth factor I receptor (IGF-IR) wherein the antiobody cimprises a light chain that uses huiman A30 VK gene, inhibits IGF-I or IGF-II binding to IGF-IR and inhibits tumor growth in vivo. The invention also discloses a corresponding encoding nucleic acid molecules, transgenic animals containing these nucleic acid molecules, methods of making anti-IGF-IR antibodies and pharmaceutical compositions comprising these antibodies.
