<P>PROBLEM TO BE SOLVED: To provide a method for purifying a polypeptide (e.g., an antibody) from a composition containing the polypeptide and at least one contaminant by using the ion exchange chromatography. <P>SOLUTION: The ion exchange chromatography for binding the objective polypeptide to an ion exchange material at first conductivity and pH, subsequently washing the ion exchange material with an intermediate buffer having different conductivity or pH, or both different conductivity and pH is used. The ion exchange material is washed with a washing buffer, however the change of conductivity or pH, or both of them from the intermediate buffer to the washing buffer is reverse to the change of the conductivity or pH, or both of them performed in the process. Solely after washing with the washing buffer, the ion exchange material is prepared for objective polypeptide molecules eluted by applying an elution buffer having the conductivity or pH used in the process, conductivity or pH different from both or conductivity and pH different from both. <P>COPYRIGHT: (C)2010,JPO&INPIT |
