Provided is a method for determination of thymidine kinase (TK) activity in a biological sample comprising the steps of contacting, in a container containing a single stranded polynucleotide as a primer or template attached to a solid surface, a Basic Reaction Mixture, in a buffer, comprising the following components: a non-attached single stranded polynucleotide as a template and/or primer, a modified deoxy nucleoside as a kinase enzyme substrate, a phosphate donor, a nucleotide polymerizing enzyme, and a kinase enzyme source devoid of TK activity, with the biological sample, incubating the mixture, optionally washing the incubated mixture to remove non-attached reaction components, determining the amount of modified deoxy nucleoside that has been incorporated into the solid surface-attached primer or template and 15 determining the TK activity present in the biological sample from the amount of incorporated modified deoxy nucleoside. Further provided are corresponding assay kits and methods of diagnosis. |
