SPECIES IDENTIFICATION ANALYSIS METHOD FOR PROTARGOL-STAINED UNCULTURED SINGLE CILIATE CELL

PURPOSE: A method for analyzing species identification of single ciliate using protargol is provided to accurately identify species by performing morphological analysis and base sequencing analysis. CONSTITUTION: A method for analyzing species identification of single ciliate cell without culturing comprises: a step of dyeing a sample containing the single ciliate cell with protargol; a step of extracting DNA from the sample and analyzing sequence; and a step of determining species. The species identification analysis comprises: a step of separating individual from a sample; a step of fixing the individual with HgCl_2; a step of washing with distilled water and treating with protargol; a step of treating with dye fixing liquid; a step of observing the individual with a microscope; and a step of isolating DNA form the individual.
