PURPOSE: A method for calculating enzyme reaction rate and a method for determining enzyme amount in a sample and computer program product are provided to accurately and efficiently calculate enzyme reaction rate. CONSTITUTION: A method for calculating enzyme reaction rate comprises: a step of obtaining optical signal according to reaction time of enzyme and substrate; a step of dividing optical signal of a sub-group based on reaction time interval; a step of calculating gradient based on time with respect to optical signal;; a step of obtaining gradient frequency to each sub group; a step of obtaining difference value of gradient value and gradient value of each sub group by reducing the gradient value of each sub group(Sn) from gradient value corresponding to minimum value; a step of adding difference value to sequential sub groups of certain number and selecting sequential sub group section corresponding to minimum value; and a step of determining enzyme reaction rate.
