<P>PROBLEM TO BE SOLVED: To provide an immunological reagent having the anti-CEA-binding properties of mouse Class III and anti-CEA mAb(monoclonal antibody) and human mAb's immunogenicity in human patients. <P>SOLUTION: A humanized monoclonal antibody has a parent mouse Class III transplanted into a heterologous antibody's framework region and an anti-CEA monoclonal antibody's complementary determination region; wherein although the humanized antibody keeps the binding specificity of the parent mouse monoclonal antibody, its immunogenicity is smaller than that of the parent mouse monoclonal antibody in a heterologous host. Preferable mouse Class III and the anti-CEA monoclonal antibody are each an MN14 antibody, while a preferable heterologous antibody is a human-derived one. A DNA construct and a vector which produce this humanized monoclonal antibody, and a diagnostic and curative conjugates each using the humanized monoclonal antibody are also provided. <P>COPYRIGHT: (C)2010,JPO&INPIT |
