PROBLEM TO BE SOLVED: To provide Apo-2 ligand variant polypeptides. SOLUTION: Apo-2 ligand/TRAIL variant containing one or more amino acid substitutions in a natural sequence of Apo-2 ligand/TRAIL is provided. The Apo-2 ligand/TRAIL variant may contain cysteine substitution. Apo-2 ligand/TRAIL variant polypeptide is included and conjugates with or bind to one or more polyol groups such as a poly(ethylene glycol) 2000 and the like. Nucleic acid molecules encoding such Apo-2 ligand/TRAIL variant, and vectors and host cells containing the nucleic acid molecules encoding the Apo-2 ligand/TRAIL variant are also provided.
