B CELL-DERIVED IPS CELLS AND APPLICATION THEREOF

Disclosed are iPS cells derived from B cells by a simple technique, technology to inexpensively provide human antibodies using said iPS cells, and mice, etc. that have been immunologically humanized using cells differentiated from said iPS cells. The cells have been cloned by bringing a nuclear reprogramming factor that does not contain a C/EBPa and Pax5 expression-inhibiting substance, especially nucleic acids that code Oct3/4, Sox2, Klf4, and c-Myc, in contact with B-cells, which cells (B-iPS cells) have undergone immunoglobulin gene rearrangement, and are pluripotent and autonomously replicating. A method for producing monoclonal antibodies against a specified antigen is characterized by collecting antibodies from a culture of B cells obtained by differentiating B-iPS cells derived from B cells immunized with said specified antigen. A method for producing immunologically humanized mice is characterized by transplanting human hematopoietic and immune system cells, obtained by differentiating B-iPS cells, into immunodeficient mice.
