PROBLEM TO BE SOLVED: To provide a metal ion sensor which detects and quantifies a protein which is synthesized in a large scale by synchronizing data of presence of metal ions with data of protein expression. Ž
SOLUTION: In the metal ion sensor, a DNA-binding domain of a heat-shock protein is bound to a peptide that forms a three-stranded coiled coil structure in response to binding of a metal ion, thereby inducing binding of the peptide to a specific sequence of DNA in the presence of metal ions. Here, aspartic acid is introduced into the peptide sequence so as to produce a marked difference in its DNA binding activity depending on the presence or absence of metal ions.
