PURPOSE: A 2-amino-5-methylimidazole derivative having substituted 4-site is provided to strongly suppress NHE-1(sodium-hydrogen exchanger-1) and to prevent and treat ischemic heart diseases. CONSTITUTION: A 2-amino-5-methylimidazole derivative having substituted 4-site is denoted by chemical formula 1. A method for manufacturing the 2-amino-5-methylimidazole derivative comprises: a step of reacting a carboxylic compound(2) with N,O-dimethylhydroxylamine under the presence of a condensing agent to obtain methoxymethylamide(Weinreb amide) compound(3); a step of reacting the methoxymethylamide compound(3) with ethylmagnesium bromide to obtain propion comound(4) having substituted 1-site; a step of performing alpha-bromination of the propion compound(4) to obtain 2-bromopropanone compound(5); a step of reacting the 2-bromopropanone compound with 1-acetylguanidine to obtain 2-(N-acetyl)amino-5-methylimidazole compound(6) having substituted 4-site; and a step of performing deacetylation of the 2-(N-acetyl)amino-5-methylimidazole compound(6) having substituted 4-site.
