<P>PROBLEM TO BE SOLVED: To provide a method which improves accuracy of an intermolecular interaction analysis in a fluorescence cross correlation spectroscopy (FCCS), without altering its optics, and to provide a program used for the method. Ž<P>SOLUTION: The method for optimizing the intensity of exciting light in the FCCS using a first exciting light and a second exciting light which have mutually different wavelengths includes: a step (a) of preparing a sample containing a first target molecule labeled, by using a first fluorescent substance which can be caused to fluoresce by the first exciting light, and a second target molecule labeled, by using a second fluorescent substance which can be caused to fluoresce by the second exciting light; a step (b) of determining two or more combinations of exciting light intensities of the first exciting light and the second exciting light; a step (c) of calculating apparent binding rates of the first target molecule and the second target molecule for all combinations determined in step (b); and a step (d) of optimizing the combination of the exciting light intensity of the first exciting light and the second exciting light, based on the apparent binding rates calculated in step (c).
