PROBLEM TO BE SOLVED: To improve detection sensitivity by performing optical detection in a state that a fluorescent label is close to a sensor in a method for detecting the quantity of a substance to be tested. SOLUTION: The detection method includes the steps of feeding a sample S to the sensor 14 on a sensor chip 10, generating an optical electric-field D reinforced on the sensor 14 by irradiating the sample S with excitation light Lo, exciting the fluorescent label F, and detecting the quantity of a substance A to be tested from the quantity of light Lf generated by the excitation. The detection device includes the sensor 14 with either electric charge on the surface thereof, and a fluorescent material F (fluorescent label) with the other electric charge containing a plurality of fluorochrome molecules f by a translucent material 16 transmitting fluorescence produced from the plurality of fluorochrome molecules f.
