PROBLEM TO BE SOLVED: To provide a method for preparing a compound that develops color by oxidation for use in measuring an organism component. SOLUTION: The method of preparing the compound that develops color by oxidation, represented by formula 3 (wherein R1, R2, R3, R4and R5are each independently a hydrogen atom, or a sulfonic acid group; and at least one of R1, R2, R3, R4and R5is a sulfonic acid group), or its salt comprises, upon introducing an amino group thereinto, isolating the resulting nitroso compound subsequent to nitrosation and further subjecting the nitroso compound to catalytic reduction with hydrogen.
