<P>PROBLEM TO BE SOLVED: To provide a therapeutic compound and method for treating a host infected with flavivirus. <P>SOLUTION: A method of inhibiting replication of a flavivirus in animal cells, and an oligonucleotide compound for use in the method are provided. The oligonucleotide analog (i) includes nuclease-resistant backbone, (ii) can be uptaken by cells, (iii) includes 8-40 nucleotide bases, and (iv) includes a sequence of at least 8 bases complementary to the region of the virus' positive strand RNA genome that includes at least a portion of a specific sequence. Exposure of cells infected with a flavivirus to the analog is effective to form within the cells, a heteroduplex structure comprising the virus ssRNA and the oligonucleotide, the heteroduplex structure being characterized by a Tm of a dissociation temperature of at least 45°C. <P>COPYRIGHT: (C)2010,JPO&INPIT |
